Maybe you need to run Notepad as administrator? An alternate to Notepad would be Hosts File Editor which is visual like Notepad but it offers a much nicer UI for editing host entries. I can open the file in Notepad OK but when I try to save it after making changes it says "Cannot create the C:\Windows\System32\drivers\etc\hosts file. Then from NotePad open the MySQL my.ini file. For example, when I manually open up notepad, then have a robot save it to a restricted network location for which I have write access, then the file saves successfully.
